I'm not about to pull that ladder up behind me. Decoding the microbiome is one of the hardest problems in biology, and we're going after it. We won $50k in rent at LabCentral, and feedback and guidance from Sanofi. Sanofi's challenge this year focused on disease modeling for more precision therapeutics: organoids, organ-on-chip, human-relevant models, and AI/ML approaches that de-risk candidates before the clinic. Outpost's Lab-in-the-Loop platform combines high-throughput experiments with machine learning in a continuous cycle to predict how the microbiome shapes drug response. This was extra special for us as this award landed during our quarterly Home Week when our global team meets in London or Boston, so all of us got to be in the room together to celebrate. Huge thanks to Karen Chandross, PhD, Zhaoling Meng, Biljana D., Laura Chambre, PhD and Dinesh Bangari from Sanofi, and to Maggie O'Toole and the broader LabCentral team and community for hosting the reception and Outpost’s lab.

We shared Outpost's Lab-in-the-Loop platform, which combines high-throughput screening of microbiome ecosystems with machine learning to understand how distinct microbiomes interact with and transform the molecules they encounter. This platform enables us to generate predictive insights into microbiome function at scale, helping us move beyond describing microbial communities to predicting what they do. Our mission to demystify the microbiome and harness its potential to improve human health resonated with the audience and we gained a lot of insights from attendees. Heidi Arjes, PhD, Kaitlyn Barrack, Ph.D., and Diego Aguirre all knocked it out of the park presenting our work and engaging with attendees. Many thanks to the amazing organizers of BBM (which is entirely trainee-led!) who always put on a stellar event. 👏

The response floored us, both the volume of questions and how sharp they were. It really feels like the right moment for the models we're building (watch this space for a big announcement soon!). What seemed to resonate most was our Lab-in-the-Loop approach. Outpost pairs high-throughput experiments with ML to learn how the microbiome and compounds interact, then validates it back in the wet lab. Kaitlyn Barrack, Ph.D. and Diego Aguirre are also presenting a poster on the same work tomorrow. Heidi, you were fantastic. Proud to build alongside you and the whole wet lab team. Onwards from here!

Joey Gardner reposted thisJoey Gardner reposted thisJust out on the Outpost blog: how much more functional signal can you actually pull out of commodity 16S sequencing? 16S tells you who's in a microbiome sample. Shotgun metagenomics tells you what they can do. The second is the readout most teams want, and the one they can least afford to run at scale. So Saif and our team tested a simple idea: pair 16S with a gut-specific genome reference (UHGG, 4,744 species, most with no cultured representative) and read enzyme function directly off the closest gut genomes. We call it UHGG-EC. Then we spent most of our effort trying to fool the result. Against paired shotgun truth for 113 human stool samples, UHGG-EC recovers more functional enzymes than the standard tool, and the gain holds up after controlling for annotation artifacts. The win isn't in the conserved core every gut shares. It's in the rare, sample-specific tail, which is exactly where discovery hypotheses come from. It's a recall-first discovery screen, not a replacement for shotgun, and we're clear about where it breaks. I built a Bayesian hierarchical model of protein melting points live in 27 minutes using Cursor and marimo. The agent was fast. But it was fast for me in a way it would have been merely noisy for someone who lacked the context. Every prompt hid a decision. To pick a protein, I had to know that a melting curve only means something when the signal has real dynamic range. To fit the curve, the four-parameter logistic's bounds had to land at exactly 1.0 and 0.0. That is dataset-specific domain knowledge. Strip the expertise out and the prompts collapse. I caught what the agent missed. The temperature column was missing from the dataframe. The agent was perfectly happy to charge ahead and fit curves to nothing. I had to send it hunting for the actual temperatures. Know your vibe zone. Go fast inside your zone of expertise; slow way down outside it. Trust scales with your ability to verify. I could trust the agent because I could check its output. The gap widens. The agent hands a loaded instrument to whoever is holding it. In expert hands it is a microscope. In novice hands it is a confocal microscope pointed at the wrong slide, producing beautiful, expensive, wrong images. Your investment compounds now instead of depreciating. Six months ago, I facilitated a Lego® Serious Play® session with the Outpost Bio team as they explored how they wanted to work together. Last week, we picked up where we left off, not by starting again, but by asking: "What must never be lost as we grow?" The answers didn't come from me. They came from the team. Through stories, metaphor and conversation, they described a culture built on trust, challenge, psychological safety and high standards. What struck me most was their conclusion that growth hadn't diluted who they are. It had strengthened them. Knowing this team, I have a sense that they will work hard to ensure this remains a strength and continues to fuel their success. Thank you, @Alex, Jenny Yang, and the whole Outpost Bio team, for your openness, curiosity, and trust. It was a privilege to continue your story with you.Joey Gardner liked thisWhat a week! Two weeks ago, the London Outpost Bio team flew into Boston, bringing our global team together for the first time since the spring. We're a company split across an ocean, London and Boston, so a week where everyone is physically together is... special! And it turns out it's the thing that matters most. In one session, we built our culture out of Lego. Scientists, engineers and commercial leaders, heads down, arguing about what a pile of plastic is supposed to represent. It sounds ridiculous. But it's not. It turns out that when you're building, you stop thinking. And this enables incredible communication and team building. Thanks, Afshan Baksh FRSA, for your magic! The prompt of that session was simple: what must never be lost as we grow? Neythen built a bridge between our two offices and made the point that connection isn't automatic; it takes active, intentional effort, especially as we scale. Samuel built three roles: wet lab, dry lab, and ops, all pulling the same load with everyone's hands on the rope. Team member after team member, the same themes surfaced without anyone steering them: psychological safety, high standards, and the courage to always share feedback with each other, even if it's awkward. When we asked what we've BECOME over the last six months, the words that came back from the builds were resilient, communicative, capable. We've grown fast and taken on more than we thought we could, and the usual story is that growth dilutes what made a place special. The team's own read was the opposite: our growth has added capability without diluting our identity. This is what it's like to build at Outpost. I've spent most of my career thinking about how teams actually work, and I can't overstate how much of a joy it is to watch a team define its own culture instead of having it handed to them on a slide. Culture isn't the values written on a wall. It's the quality of the space we create between one another, built one interaction at a time. Reflecting, challenging, laughing, building the thing together. Huge thanks to Jenny for being my partner in crime, building this company together. And to Afshan Baksh FRSA and the Transformation Space Ltd team for guiding the session, and to our team who brought their whole self to the week. Our best days are still ahead! Oh... and we won a $50k golden ticket from Sanofi in the middle of our home week. What a thrill!! Onwards from here! 🚀
