Biplab Das posted thisRecent review -In Vitro and In Vivo Metabolism Studies -Biplab Das and Chandra Prakash published in Drug Discovery and Evaluation : Safety and Pharmacokinetic Assay (2023) Despite major strides in the understanding of drug metabolic processes, knowledge in several areas is still lacking, and predictive tools, while significantly improved, need further enhancement. In silico tools are improving with better ability to predict labile metabolic sites. The lack of available data in humans in early drug discovery stages makes it difficult to have well validated, rapid, in vitro ADME tools, in complement with relevant in vivo preclinical animal models that can accurately predict the ADME and safety of an NME in humans. In recent years, artificial intelligence (AI) and machine learning (ML) have also emerged as an indispensable tool in preclinical discovery and ADME/Tox to help teams make the right decision. Taken together, drug metabolism as a discipline is a critical component in drug discovery and development, contributing significantly to the process of identifying new drug molecules and bringing them to patients with unmet medical needs. This chapter describes, in detail, the utility and the challenges of these tools both with in vitro and in vivo metabolism studies that are used to design better molecules that have properties that include lower systemic clearance, and also assist in the selection of nonclinical species for use in safety assessment studies in preclinical development. Additionally, these tools aid in the understanding of reactive metabolites to avoid idiosyncratic adverse drug reactions. Practical case examples are provided.

Biplab Das liked thisBiplab Das liked thisAfter more than three decades in drug discovery and development, I am stepping into retirement with a full heart, deep gratitude, and genuine excitement for what lies ahead. I feel incredibly fortunate to have spent my career doing work that I truly loved, alongside people who inspired me every day. My time at Bristol Myers Squibb and Agios Pharmaceuticals shaped me not only as a scientist and leader, but as a lifelong learner. I had the privilege of contributing to programs that moved from early discovery to patients, but even more importantly, I was part of teams that believed deeply in collaboration, rigor, and purpose. I owe so much to my mentors, who challenged me, trusted me, and generously shared their wisdom over the years. I am equally grateful to the many colleagues and cross‑functional partners around the world who taught me something new every day. Science is never a solo endeavor, and I have been lucky to work with people who made even the hardest problems feel solvable—together. One of the greatest joys of my career has been the opportunity to build teams and invest in the growth of junior scientists. Watching them develop confidence, curiosity, and scientific judgment—and then go on to do amazing things of their own—has been deeply rewarding and something I will always cherish. Later in my career, gaining a CRO and integrated drug discovery perspective from Syngene international, including building end‑to‑end discovery capabilities across the US and India, broadened my view and reinforced for me the importance of integration and partnership with academia and never stopping learning. As I begin this next chapter, I’m looking forward to slowing down just enough to savor it. I plan on giving back through volunteering; staying intellectually engaged and connected to the scientific community in thoughtful ways; spending lots of precious time with my granddaughter and finally making space to learn new skills that I never quite found time for during a wonderfully full career. Though this chapter is coming to a close, the relationships and friendships endure. I hope to stay connected and would be delighted to hear from you as our paths continue to cross in new ways. Thank you to everyone who has been part of my journey—you made this career far more meaningful than I ever imagined. Here’s to lifelong learning, new beginnings, and a little more time for what matters most.

Ultrasmall gold nanoclusters enable more sensitive imaging of the kidneys compared to iodinated contrast which we hope translates into less risk of contrast induced nephropathy. Importantly, differences in renal pharmacokinetics with the gold nanoclusters demonstrated disease mechanism-specific imaging signatures and distinct time-attenuation behaviors that allowed for the noninvasive characterization of different types of kidney injury.
