In this study, researchers compared five CSF immunoassays for measuring Beta-Amyloid (1-42) against both an antibody-independent mass spectrometry reference method and visual amyloid PET assessment, in a cohort of 262 patients with mild cognitive impairment or subjective cognitive decline. A key finding was that while newer assays correlated more closely with the mass spectrometry reference, the classic INNOTEST assay showed stronger concordance with visual PET classification, and its signal was partially quenched when CSF samples were spiked with recombinant Aβ1-40 peptide. Importantly, the accuracy of the newer assays improved significantly when Aβ42:Aβ40 or Aβ42:tau ratios were used rather than Aβ42 levels alone, achieving performance comparable to the classic assay. In this Nature Neuroscience study, researchers investigated whether phosphorylated Tau can impair neurovascular function independently of neurodegeneration, a question with important implications for understanding the earliest stages of tauopathy. Using two transgenic mouse models expressing mutant Tau, the team found that neurovascular coupling, the mechanism by which neural activity drives increases in cerebral blood flow, was selectively suppressed before the onset of tau pathology, neuronal loss, or cognitive decline. Mechanistically, the researchers identified that tau disrupts the interaction between neuronal nitric oxide synthase (nNOS) and the postsynaptic scaffold protein PSD95, reducing nitric oxide production during glutamatergic synaptic activity. Since nitric oxide is a key mediator of activity-dependent cerebrovascular dilation, its deficiency provides a direct link between tau accumulation and impaired cerebral blood flow. Notably, suppressing tau production reversed these neurovascular deficits, supporting tau as a tractable therapeutic target at the vascular level. rPeptide's Tau-441 P301L Mutant (cat# T-1014) was used in cerebral blood flow measurements and two-photon and laser speckle imaging experiments throughout this work. New research from UCLA links long-term exposure to chlorpyrifos, a widely used agricultural pesticide, to more than a 2.5-fold increase in Parkinson's Disease risk. Beyond the epidemiological findings, the study went further to uncover why: chlorpyrifos disrupts autophagy, the cell's internal cleanup system, allowing toxic Alpha-Synuclein to accumulate in dopamine-producing neurons. In this Nature Neuroscience study, researchers successfully engrafted human microglia derived from embryonic stem cells into the mouse brain, creating a chimeric model designed to address a significant limitation in Alzheimer's Disease research: roughly one-third of putative Alzheimer's Disease risk genes lack adequate mouse orthologs. By transplanting human stem cell-derived microglia into a humanized mouse model, the team was able to study human-specific microglial responses that would otherwise be inaccessible in standard animal models. The engrafted human microglia closely recapitulated the transcriptional profile of primary human microglia isolated ex vivo and showed expression of human-specific Alzheimer's Disease risk genes.
