About us
Our mission is to develop groundbreaking therapies to reverse mitochondrial dysfunction by leveraging a deep mechanistic understanding of mitochondrial biology. Mitochondria are essential energy-converting cellular organelles found in nearly every cell type throughout the body. Dysfunctional mitochondria are involved in more than 50 diseases. The most severe of these are broadly termed mitochondrial diseases, a group of rare genetic conditions which affect individuals of all ages. Mitochondrial dysfunction also plays an important role in more common diseases, including aging-related disorders. At Pretzel, we are developing first-in-class therapeutics that can address mitochondrial dysfunction at its roots. By taking aim at the genetic drivers of mitochondrial dysfunction, we believe our approach could be effective across a wide range of diseases which lack meaningful treatment options today.

Euromit 2026, the leading international congress dedicated to mitochondrial disease research, held last week was a great opportunity for the Pretzel team to connect and share our latest research with scientists and clinicians across the globe. We provided an update on our lead program PX578, a first-in-class POLG activator for the treatment of mitochondrial DNA depletion syndromes; discussed our therapeutic targeting of SAMHD1 to restore mitochondria function in mtDNA depletion models; and participated in discussions during the Therapeutic & Diagnostic Developments session. Looking forward to more exciting progress on many fronts in the months to come!

PharmaSource spoke to Baruch Harris, COO at Pretzel Therapeutics, about how Pretzel has kept its mitochondrial biology pipeline moving through a brutal bear market, including a $72.5M Series A, a Series B, and the acquisition of Rome Therapeutics using stock, not cash, to add balance sheet strength without a dilutive VC round. The financing strategies he outlines go well beyond "go raise more money." Spinouts to unlock hidden portfolio value. Non-dilutive grants from NIH, BARDA, and disease foundations like Parkinson's UK. And when it comes to deciding where to spend what you have, his framework is to go programme by programme and ask what it will cost to reach the next value inflection point, and whether that data will actually attract a partner or the next round. If the answer is no, pause it, even if the science is genuinely interesting. "The more your lead program looks like a medicine, the better it is for continuing to get the capital you need."
