The molecule you produce is the molecule your biology has to work with. RNA quality isn't just a manufacturing consideration. It's a biological one. RNA degradation and dsRNA contamination can introduce variables that downstream development must account for. For the applications pushing RNA biology forward, this matters most: Complex RNA constructs need to arrive complete. A truncated molecule may not function as intended because key biological mechanisms depend on the full sequence being intact. Personalized cancer vaccines are patient-specific by definition. RNA is one of the most powerful molecules in biology, and one of the most fragile. From the moment it's transcribed, RNA is vulnerable. IVT chemistry, shear forces from handling, and time in solution all work against it. The longer a construct is, the more surface area there is for something to go wrong. This is the scientific reality NTx Bio built around. Nature Reviews Drug Discovery just published an in-depth look at what it will take to get mRNA therapeutics from vaccines to a broader class of treatments, and manufacturing quality is one of the central challenges. The review covers a decade of clinical progress across enzyme replacement therapies, personalized cancer immunotherapies, genome-editing approaches, and immune cell reprogramming via CAR-T. The clinical momentum is real, with candidates from Moderna, BioNTech, Intellia, and Beam Therapeutics advancing into Phase II and III trials. But the authors are clear that mRNA purity, dsRNA contamination, and scalable production remain barriers that have to be solved for mRNA therapeutics 2.0 to deliver on its promise. For applications like enzyme replacement and personalized neoantigen vaccines, the manufacturing platform isn't a downstream consideration, it's a gating factor. These aren't abstract problems. A recent opinion piece in The New York Times highlights a pivotal shift in medicine. Written by Jeff Coller, PhD, who directs the RNA Innovation Center at Johns Hopkins University and serves on NTx’s Scientific Advisory Board, the article reflects on a child born with a severe rare disease who was treated with a personalized CRISPR-based therapy, designed and delivered in a matter of months. Today, he’s thriving. The science is no longer the limiting factor. Advances in gene editing and mRNA have made it possible to develop highly targeted, patient-specific therapies, yet these breakthroughs remain the exception, not the standard. The constraint is infrastructure. If gene editing is to function like molecular surgery, manufacturing must evolve to match. It must be fast, flexible, and patient specific. Traditional large batch biomanufacturing was not designed for this paradigm. NTx Bio is focused on enabling rapid, automated, small batch RNA manufacturing to support the transition from one off breakthroughs to scalable clinical practice.
