#ATS2026 reinforced the innovation that has taken hold in the respiratory space. We were proud to present late-breaking Phase 1 data for ALX1 this week in Orlando. Across the meeting, there was clear momentum behind new modalities targeting epithelial-driven inflammation, growing adoption of "endotyping" to match therapies to the right patients, new approvals in pulmonary fibrosis and severe asthma, and a strong “Suspect Bronchiectasis” campaign to build awareness around bronchiectasis – a rapidly growing and underdiagnosed disease in the U.S. A big congratulations as well to the Avalyn Pharma team on their recent IPO success, a strong validation for advancing inhaled nebulized therapies. Today at #ATS2026, our CMO, Paul Bruinenberg MD MBA presented late-breaking Phase 1 data for ALX1, our first-in-class inhaled nitric oxide (NO) prodrug designed for targeted delivery to the lungs. The data showed inhaled ALX1 was safe and well tolerated across all dose levels evaluated, with no dose-limiting toxicities, no clinically significant methemoglobinemia, and no bronchospasm observed in healthy volunteers – even with successful escalation above the calculated human effective dose. The study also demonstrated dose-proportional pharmacokinetics, supporting continued development in bronchiectasis, COPD, and other muco-obstructive airway diseases. We’re thrilled to share that Vast Therapeutics has reached a meaningful milestone in our journey to improve outcomes for patients with serious lung diseases. We have successfully completed the Single Ascending Dose (SAD) portion of our Phase 1 clinical program for ALX1, the first-in-class, inhaled pro-drug designed to deliver localized nitric oxide deep into the lung. Key highlights: - 32 healthy adult participants across four escalating dose levels - ALX1 was well tolerated with no severe or serious adverse events - Dose escalation achieved across all four pre-specified levels This achievement validates our hypothesis that a portable inhalation device delivering nitric oxide in liquid form may overcome prior limitations of inhaled gas, setting the stage for meaningful therapeutic impact in conditions like bronchiectasis and cystic fibrosis. (As noted by Professor James Chalmers from the University of Dundee, the mechanism holds significant promise.) With the Multiple Ascending Dose (MAD) study already underway, we’re targeting full Phase 1 read-out in Q1 2026 and initiation of our Phase 2 patient program in the first half of next year.
