Excited to share our newest preprint, the result of a fantastic collaboration with Harvey Tian and Adam Bisogni at Inso Biosciences. In this work, we show how micro-pillar technology enables Nanopore sequencing libraries in ≤45 minutes, dramatically shortening the path from sample to answer. Using this approach, we successfully generated libraries from whole blood, LCLs, tissue, and even a single drop of blood from a finger prick. Most excitingly, we demonstrate that brain tumor classification using methylation alone can be achieved with ~10 minutes of sequencing—or roughly 1 hour from the start of library prep to result. I’ve been looking forward to sharing this story for a few reasons: - Sample-to-answer, fast: It shows that rapid molecular diagnostics are possible with minimal hands-on time, limited technical expertise, and without expensive automation or robotics. - Point-of-care potential: This provides a clear path toward point-of-care long-read sequencing, something we need to start preparing for now—clinically, logistically, and ethically (counseling, interpretation, reporting). - Equity and access: The workflow highlights how emerging sequencing technologies could be adapted for low-resource or decentralized settings, enabling fast, actionable diagnostics where they’re needed most.
